5-Amino-1MQ is a small-molecule research chemical, not a peptide, studied entirely in cell and animal models. There are no published human trials. This page describes the animal findings behind its reputation, not established benefits.

Overview

5-Amino-1MQ has no demonstrated benefits in humans — every claim made for it traces to mouse experiments. What those experiments showed is genuinely interesting: obese mice given this NNMT (nicotinamide N-methyltransferase) inhibitor lost weight without eating less, and aged mice gained measurable muscle strength. But the compound has skipped the entire clinical pipeline between "promising in mice" and "works in people," and it is sold online as if that gap did not exist. Below is what the actual studies found, with their numbers and limits.

The mouse obesity study everyone quotes

The study that put 5-Amino-1MQ on the map treated diet-induced obese mice with the compound at 20 mg/kg, injected subcutaneously three times a day for 11 days (Biochemical Pharmacology 2018). Treated mice, compared with saline controls:

  • Lost a significant amount of body weight — while food intake did not change, suggesting a metabolic mechanism rather than appetite suppression.
  • Had smaller fat pads and markedly reduced adipocyte (fat-cell) size.
  • Showed lower total plasma cholesterol.
  • Showed increased NAD+ and S-adenosylmethionine inside cells, consistent with the intended mechanism.

Worth noting: that dosing schedule — weight-based injections three times daily — bears no resemblance to the once-daily oral capsules sold to consumers.

Muscle function in aged mice

A second research thread involves aging muscle. In 24-month-old mice (roughly equivalent to elderly humans), two months of NNMT-inhibitor treatment increased grip strength by about 40% versus sedentary controls — more than exercise alone (~20%) — and combining the inhibitor with exercise produced a ~60% gain, along with larger muscle-fiber cross-sections (Scientific Reports 2024). This is the study behind "5-Amino-1MQ for muscle" marketing. It is one experiment, in aged mice, with no human counterpart — and its authors present the result as a case for developing therapeutics, not as evidence that any product sold today works.

Why NNMT is the target

The rationale predates the compound. In 2014, researchers showed that knocking down NNMT expression in fat and liver protected mice from diet-induced obesity by increasing cellular energy expenditure — NNMT normally consumes both a methyl donor (SAM) and nicotinamide, the precursor the cell uses to rebuild NAD+ (Nature 2014). Elevated NNMT in fat tissue tracks with metabolic dysfunction, so inhibiting it is a legitimate scientific hypothesis — one that academic and industry groups continue to pursue with newer NNMT inhibitors. 5-Amino-1MQ is simply one early tool molecule built to test that hypothesis in animals, not the refined end product of that research program.

The missing step — humans

Between a mouse result and a human benefit stand pharmacokinetics, dose finding, safety trials, and efficacy trials — none of which exist for 5-Amino-1MQ. No published human study has measured whether oral capsules even achieve meaningful blood levels, let alone whether they change weight or muscle. Until that work happens, "benefits" is the wrong word; the accurate phrase is promising mouse pharmacology.
