Overview
AICAR (5-aminoimidazole-4-carboxamide ribonucleotide) is a nucleoside analog that, once inside cells, activates AMP-activated protein kinase (AMPK) — a master regulator of cellular energy. Because AMPK is normally engaged by exercise and energy stress, interest in AICAR centers on:
- Reproducing some of the metabolic signals associated with physical activity.
- Effects on glucose uptake and fatty-acid oxidation in muscle.
- Signaling linked to mitochondrial biogenesis and oxidative capacity.
These are the mechanisms researchers study; they are not a promise of any specific athletic, metabolic, or cosmetic result in humans.
AMPK activation and metabolism
In cell and animal models, AICAR activates AMPK and shifts metabolism toward energy production rather than storage. This has driven interest in glucose handling and fat oxidation. The effects, however, are broad and systemic because AMPK sits at the hub of many pathways.

These findings generated excitement but also skepticism about whether comparable effects occur in humans and at what cost in delivery and exposure.
How AICAR is framed
AICAR is often discussed alongside other metabolic modulators, but it differs in important ways:
- It is a small molecule, not a peptide, despite catalog grouping.
- It acts on AMPK broadly rather than on a single hormone receptor.
- It occurs naturally as a purine-pathway intermediate, complicating detection questions.
Evidence and caveats
