Researchers at UCLA and elsewhere synthesized short fragments of it — called granulysin-derived peptides, or GDPs — and showed that fragments carrying a helix-loop-helix motif killed Cutibacterium (formerly Propionibacterium) acnes in vitro, killed it inside isolated human microcomedones, and suppressed the cytokine release that C. acnes provokes (McInturff et al., J. Invest. Dermatol., 2005). The stated motivation was antibiotic resistance: the peptides were bactericidal, which the authors framed as an advantage over bacteriostatic antibiotics.
Is the research on "Oligopeptide-10" specifically? This is the distinction that most ingredient write-ups skip. The published literature studies granulysin-derived peptides identified by laboratory codes — GDP, peptide 31-50v44w, GDP-20 — not an ingredient named Oligopeptide-10. The INCI name is a labeling designation, and no peer-reviewed trial has been published under it. Reading the GDP literature as evidence for a product labeled Oligopeptide-10 requires assuming the materials are equivalent, which the papers do not establish.
What did the human GDP studies actually find? Two, both small and neither placebo-controlled. Lim and colleagues screened 20 synthesized GDPs for antimicrobial activity, then had 30 patients with facial acne apply a topical formulation containing 50 ppm of the best-performing peptide twice daily for 12 weeks: pustule counts and Korean Acne Grading System scores fell significantly, but comedone counts rose, and there was no control group (Int. J. Dermatol., 2015). In a 60-patient Russian study, adding topical GDP-20 to low-dose systemic isotretinoin produced substantial lesion reduction over 12 weeks — but both arms received isotretinoin, a potent approved drug, so the peptide's own contribution cannot be read off the result (J. Cosmet. Dermatol., 2022).
Is it a substitute for acne medication? No, and the difference is legal as well as clinical. The FDA defines a drug partly by intended use — "articles intended for use in the diagnosis, cure, mitigation, treatment, or prevention of disease" — so a product marketed to treat acne is a drug regardless of how it is packaged (FDA, Is It a Cosmetic, a Drug, or Both?). Cosmetic ingredients other than color additives need no FDA approval before marketing (FDA). A cosmetic peptide sold for the appearance of clearer skin is making a deliberately narrower claim than benzoyl peroxide or a prescription retinoid.
Who funded the underlying research? Worth knowing before weighing it. The 2005 granulysin-peptide paper carries a conflict-of-interest statement noting that several authors were employees, consultants, or scientific advisory board members of Ansata Therapeutics, the company developing the peptides (J. Invest. Dermatol., 2005). That is normal for translational work and does not invalidate it, but it means the ingredient's foundational data were produced by parties with a commercial stake, and no independent group has replicated the clinical findings.
